常温纤维素降解菌群的筛选及其特性初探

    Screening and Characteristics of Cellulose Degrading Microbial Flora Under a Constant Temperature

    • 摘要: 在改良PCS培养基的基础上,通过定期改变传代方法,从污泥、森林土、发酵腐熟牛粪、堆肥处理麦秆中筛选出1组纤维素分解能力强而稳定的纤维素混合菌群(代号为Xc),100 mL培养物在37℃静置培养5 d,对滤纸纤维素、脱脂棉、麦秆(片)、牛粪纤维降解率分别达到81%、41%、25%、40%。Xc在37、28℃分解滤纸时,分别在第5天、第7天时CMC酶活性最高,分别达到104.5、95.3 U.mL-1。在初始pH5~9的培养基上接种均能表现出较强分解能力。采用变性梯度胶电泳(DGGE)法比较来源于不同时期菌群16S rDNA条带表明,菌群结构稳定。

       

      Abstract: A group of cellulose degrading microbial flora (Xc) was screened from sullage,forest soil,cow manure and composted wheat straws with the technology of regularly altering the regeneration method during incubation on the modified PCS culture medium.The flora showed a stable and strong capability of degrading cellulose.About 81%,41%,25% and 40% of the cellulose in filter paper,absorbent cotton,wheat straw and cow manure,respectively,was degraded with 100 mL of Xc at 37 ℃ in 120 hours under static culture.In decomposing filter paper,its CMC enzyme activity peaked up to 104.5 and 95.3 U·mL-1,on D5 and D7,at 37 and 28 ℃,respectively.Xc could be inoculated into culture media ranging in pH from 5 to 9,however,in the end of incubation the final pH of the medium tended to be neutral.The Xc cellulose-decomposing activity may last more than 22 days as long as supply of substrates continues.Xc is very stable in structure.Its main DNA bands remained unchanged as is demonstrated by comparison of 16S rDNA with the DGGE method of Xc from different time periods of the culture.
