江苏沿海斑点叉尾鮰养殖环境中抗生素抗性基因污染特征及其与环境因子相关性

Pollution Characteristics of Antibiotic Resistance Genes and Their Correlation with Environmental Factors in the Coastal Channel Catfish (Ictalurus punctatus) Aquaculture Areas of Jiangsu Province

  • 摘要: 抗生素抗性基因(antibiotic resistance genes, ARGs)是一类新型污染物。为了解江苏沿海斑点叉尾鮰(Ictalurus punctatus, 以下简称"鮰")养殖环境中ARGs的污染现状, 采用实时荧光定量PCR技术, 对典型鮰养殖场环境样品中ARGs的绝对丰度进行检测分析, 同时探究ARGs与环境因子的相关性。结果表明, 水体样品中sul1、sul2、tetAtetMtetQermBmphAaad检出率为100%, sul3、qnrBqnrSblaCTX-M检出率为50.0%~83.3%, 不同采样点ARGs总绝对丰度为3.83×106~1.12×107 L-1, 其中丰度最高的3种ARGs为sul1、tetAtetM; 沉积物样品中磺胺类和四环素类ARGs检出率(100%)及绝对丰度(最高均达到107数量级)均较高, 其他ARGs检出率介于66.7%~100%之间。不同地区的特征性ARGs存在差别。冗余分析(RDA)结合皮尔逊(Pearson)相关性分析显示, 水体DO、NO3--N、NH4+-N浓度及ORP、EC、pH值、水温是影响水体中ARGs组成与分布的关键性因子, 而影响沉积物中ARGs组成与分布的关键性因子是沉积物TOC、TN、TP浓度及ORP。总体而言, 江苏沿海地区鮰养殖环境中ARGs组成丰富, 其中磺胺类及四环素类为主要检出ARGs。总体上沉积物中主要检出ARGs丰度高于养殖水体, ARGs分布呈现明显的地域特征, 环境因子对ARGs组成、丰度及分布影响较大。本研究可为江苏沿海地区鮰养殖环境中ARGs风险评估及防控提供基础数据支撑。

     

    Abstract: Antibiotic resistance genes (ARGs) is a new type of pollutants. In order to understand the pollution level of ARGs and its influencing factors in the coastal channel catfish (Ictalurus punctatus) aquaculture areas of Jiangsu Province, a real-time fluorescent quantitative PCR technique was used in this study. The absolute abundance of 12 kinds of ARGs in the environment samples were detected and analyzed, and their correlation with environmental factors were investigated. Results show that: (1) The detection rates of sul1, sul2, tetA, tetM, tetQ, ermB, mphA and aad in water samples were 100%, and the detection rates of sul3, qnrB, qnrS and blaCTX-M were between 50.0% and 83.3%. The total absolute abundance of ARGs in different sampling points was 3.83×106-1.12×107 copies·L-1, among which sul1, tetA and tetM were the most abundant ARGs. (2) In sediment samples, the most noteworthy ARGs were Sulfonamides and Tetracyclines because of their detection rates (100%) and absolute abundances (up to 107 orders of magnitude), and the detection rates of other ARGs were between 66.7% and 100%. (3) Characteristic ARGs in water and sediment samples vary in different regions. (4) Redundancy analysis combined with Pearson correlation analysis show that DO, ORP, EC, NO3--N, pH, WT and NH4+-N were the key factors affecting the composition and distribution of ARGs in water, while in sediments were ORP, TOC, TN and TP. Overall, the composition of ARGs is abundant, among which sulfonamides and tetracyclines are the main detected ARGs, and the abundance of ARGs in sediment is higher than that in water. The distribution of ARGs show obvious regional characteristics. Environmental factors have a great influence on the composition, abundance and distribution of ARGs. This study offers basic data support for the risk assessment and prevention and control of ARGs in the coastal area of Jiangsu Province.
